POSTDOC position
Cotranslational processing mechanisms: towards a dynamic 3D model
We are looking for a highly motivated postdoctoral research fellow to join our laboratory at the Centre National de la Recherche Scientifique located close to Paris. We are interested in removal of N-terminal methionine from newly synthesized proteins is ensured by a specialized biological system, the NME pathway. This maturation process acts early during protein synthesis, as soon as the nascent polypeptide emerges from ribosomal exit tunnel. However, although the co-translational feature of NME is known for several decades, the nature of ribosome-NME interactions is poorly documented. Moreover, action of other co-translational factors at the extremity of ribosomal exit tunnel assumes existence of strict spatiotemporal rules to ensure fine regulation of co-translational events involving ribosomes. We have recently started a vast project to understand the dynamic of co-translational events at the vicinity of ribosomes, especially by studying interactions with NME pathway. The postdoc project will allow answering some questions, and will consist to identification and characterization of macromolecular complexes involving NME pathway enzymes, by molecular biology, biochemical/biophysical, and structural biology approaches. The postdoc will take in charge all the structural aspect of the project. The person will be involved also in the preparation of the samples necessary for the different experimental conditions related to all tasks. The person should have strong expertise in i) crystallography ii) biochemistry including cloning, expression and production of proteins from different systems and iii) molecular biology. Particularly, experience in structural biology of complexes, ideally with ribosome, will be a prerequisite. For this project, we believe that an experienced autonomous person is needed.
